You can press the delete effect button but make sure the correct track is selected. (Command Palette | FX Tab) You can also enable Lift/Overwrite Segment mode, click on the clip with the effect and press delete. Be careful because if there isn't an effect on the clip, the clip will be deleted...

Autosequence automatically takes clips and puts them into a sequence at the timecode associated with the clips. You can remove locators from the timeline in two ways. Select the locator in the timeline and press delete or click the locator in Locators window and press delete.

You add locators by clicking a locator button. Depending on your settings, the locators window may appear that will allow you type comments associated with that locator.
