Autosequence automatically takes clips and puts them into a sequence at the timecode associated with the clips. Removing the black (filler) from the sequence would move the clips from their correct timecodes. A better solution might be to select all of the clips in the bin and drag them to the timeline window. You can press the delete effect button but make sure the correct track is selected. (Command Palette | FX Tab) You can also enable Lift/Overwrite Segment mode, click on the clip with the effect and press delete. Be careful because if there isn't an effect on the clip, the clip will be deleted...

You can change a sequence's start time by typing in the sequence's start column in the bin. You can also change the start time by right clicking in the record monitor and choosing Sequence Report.

The Media Creation dialog box (Project window | Settings Tab) is exactly what you need. Click the Render tab and choose the drive you want to render to. Now, when you want to render, this will be the default drive. You can also set default drives for importing, titles and motion effects.
